PAPANICOLAOU STAIN
Study of morphology of squamous epithelial cell
Principle:
Papanicolau Stains are used in conjunction with Hematoxylin nuclear stains in the diagnosis of malignant cytological disease. EA-36 and EA-50 are used in conjunction with OG-6 for gynecological staining. EA-65 is used with OG-6 for non-gynecological staining. When performed properly, the stained specimen should display hues from the entire spectrum: red, orange, yellow, green, blue, and violet. The chromatin patterns are well visible, the cells from borderline lesions are easier to interpret and the photomicrographs are better. The staining results in very transparent cells, so even thicker specimens with overlapping cells can be interpreted.

PROCEDURE:
1. Prepare a smear on clear, dry glass slide.
2. Fix with Ana fix.
3. Pour Dehydrant Reagent on slide for 3 minutes.
4. Wash with distilled water.
5. Flood the smear with Hematoxylin reagent for 1 min.
6. Add 3 drops of Phosphate Buffer which is not provided for 10 seconds.
7. Wash with Dehydrant 2 times for 30 seconds.
8. Pour Working Stain on slide. Allow it to remain for 45 seconds.
9. Wash with Dehydrant for 30 seconds.
10. Rinse with Xylene.
11. Allow the slide to air dry.
12. Mount with glass cover using a drop of DPX.
RESULTS:
When Staining with Papanicolaou cytoplasm appears with various shades of blue, pink, orange red and green.

Wright’s Stain With Buffer
Principle:

NEISSER′S METHYLENE BLUE
Staining Solution for Metachromatic staining.
Principle:
Well-developed granules of volutin (polyphosphate) may be seen in unstained wet preparations as round refractile bodies within the bacterial cytoplasm. With basic dye, they tend to stain more strongly than the rest of the bacterium, and with toluidine blue or methylene blue they stain metachromatically, a reddish purple colour. They are demonstrated most clearly by special methods, such as Albert’s and Neisser’s, which stain them dark purple but the remainder of the bacterium with a contrasting counterstain. The diphtheria bacillus gives its characteristic volutin-staining reactions best in a young culture (18-24 hours) on a blood or serum medium.

H&E STAIN
Intended Use:
HISTOPATHOLOGY STAIN.
Principal:
Alum acts as mordant and hematoxylin containing alum stains the nucleus light blue. This turns red in presence of acid, as differentiation is achieved by treating the tissue with acid solution. Bluing step converts the initial soluble red color within the nucleus to an insoluble blue color. The counterstaining is done by using eosin which imparts pink color to the cytoplasm.
